Abstract: An efficient protocol was developed for in vitro shoot regeneration from an aseptically grown seedling of Caesalpinia decapetala (Roth.) Alston using epicotyls explant on MS medium supplemented with BA (1.0 mg L-1). It induced maximum number of multiple shoots with 80% response. Further proliferation and elongation was achieved on MS medium containing combinations of BA and NAA (1.0+0.5 mg L-1). The elongated shoots were harvested and cultured on half strength MS medium containing IBA, NAA and IAA (0.5-2.0 mg L-1). NAA (1.0 mg L-1) has produced maximum number of roots per shoot. Rooted plantlets were made to acclimatize, hardened and gradually transferred to the field condition with 80% survival rate. © 2008 Society for Biology and Biotechnology.
