Serotype-5 & 8 capsular polysaccharides are predominating among clinical isolates of Staphylococcus aureus. The results of the experiments in animal models of infection have revealed that staphylococcal capsules are important in pathogenesis of S. aureus infections and S. aureus capsular antigens are surface associated, limited in antigenic specificity and highly conserved among clinical isolates. With the emergence of MDR strains of S. aureus new strategies are needed to combat staphylococcal infections. Production of a conjugate vaccine based on capsular subunit types is considered as a better option. In this regard we isolated a potent strain on the basis of capsular production under different conditions (In vitro and In vivo) for the development of whole cell and capsular sub unit vaccine against S. aureus. The rabbit immunized with Reynolds stain had lower level of antibodies titres in rabbits as measured by the tube antibody titres, than the local isolate of the same capsular serotype, when immunized with heat inactivated whole cells. The findings of this study indicated that the local potent strain is very much useful for the preparation of efficient vaccine to prevent S. aureus infections. Further work is undertaken to determine the protective efficiency of antibodies against S. aureus in the invasive and non invasive infections.en_US
